Relatively new with Trimix ( total ED after radical prostatectomy 17 months ago, 74 years old had been using tadalafil successfully before surgery) did Bimix as test injection in docs office actually was with an APRN got maybe 50-60% erection so she said you need Trimix for the alprostadil, got Rx for 30-1-10 (alprostadil is 10) started 25 units got up to 50 units erections maybe a 70% gave her a call asking for higher concentration as would be more economical and inject less volume she sent Rx for 30-4-40.
She said start at 30 units I did 25 wife said 'like old times' I didn't think was much harder maybe 75% lasted about 90 minutes I was still looking for that 'Raging Hard-on' so went up to 30 units then 35 units paradoxically erection seemed to be less intense and the ache was mounting.
Decided to experiment did 8 units, onset of action took longer but erection was as good as the 25 units and barely any ache.
May try going to 5 units of the 30-4-40 and see.
Anyone else note less is more?

It’s likely you’re not hitting the cavernosal space reliably, so getting inconsistent results.

When I was using trimix, I’d use a VED and pump up, put a constriction ring around the base, and then inject. Everything is plumped up and you really can’t miss the space at that point. Then massage your penis to get the medication distributed, remove the ring, and it’s off to the races.

My mix is 30-3-20, so about midway between your two. I started with a dose that varied between 3 and 5 units, which occasionally took me to the 4 hour point. Note that the stiffness seemed super hard regardless of the dose but the duration would vary.

I've gradually backed down to 1¼ to 1½ units and I stay hard - perhaps not super hard, but enough - for just the right amount of time. That can be between 30-60 minutes and goes 50% shift at ejaculation.

Às to proper injection, I use a ring and some type of stimulation (best is a little sucking from the wife) to plump up my dick. This makes it easier to find the cavernosum. I always inject to the hilt (8mm needle), then aspirate a little blood to confirm hitting the cavernosum, then inject the blood and TriMix. In the past, if I didn't aspirate. I would occasionally miss the spot.
